Protein Biopharmaceuticals: Probing correct folding with new near IR, polarised Raman spectroscopy, and circular dichroism spectroscopies
Simple quantitative measurements of protein structure with minimal input from users are under demand in industry for quality by design (Jameel et al., 2015). This aim of this project was to advance methods to extract protein secondary structure information from circular dichroism (CD), infrared (IR), and Raman optical activity (ROA) spectroscopies and to extend protein spectral libraries for these methods. The structure determination methods are the Secondary Structure Neural Network (SSNN) previously applied to CD, and independent component analysis (ICA) which holds the potential to isolate components which relate to secondary structure in sets of spectra.
