Abstract: Microfluidics has become an important tool in analytical bioscience. The possibility to precisely control microscale flows opens up a range of possibilities in biosensing as well as in cells and organs on a chip. During the presentation, I will discuss our work in the context of precision oncology. In particular, I will present a recently developed flow-rate independent multiscale liquid biopsy device. The simple device that integrates seamlessly in conventional laboratory workflows was used to capture circulating tumour cells (CTCs) from 72 cancer patients. We then evaluated the PD-L1 expression in 36 samples and found that 41% of the patients had PD-L1+ CTCs. I will also discuss our work in organs on chip where we use microfluidics to mimic, or perturb microphysiological conditions using flow-mediated mechanical and chemical stimuli. I will focus on a new fluidic lid compatible with 96 well plates that we are developing to interrogate the role of the tumour microenvironment in modulating response to chemo- and immunotherapy.
