Next Generation Sequencing
Library Preparation Service
The Genomics Facility offers numerous sequencing library preparation methods. We routinely produce Illumina compatible libraries using the following kits;
- New England Biolabs NEBNext Ultra Express RNA
- New England Biolabs NEBNext Ultra II FS DNA
- 16S Metagenomic Amplicon libraries
We have experience with the following kits which can be used on a per-project basis;
- Illumina TruSeq RNA V2
- Illumina Stranded Total RNA
- Illumina TruSeq DNA
- New England Biolabs NEBNext RNA
- Nugen Ovation Ultralow Library Systems (including Nugen Ovation RNA-Seq System)
- Illumina Nextera XT DNA and Nextera DNA Flex
Single-cell Libraries - 10X Chromium Controller
The Chromium Controller uses advanced microfluidics to perform single cell partitioning and barcoding. Powered by Next GEM technology, the Chromium Controller enables integrated analysis of single cells at massive scale.
If you would like any further details on the above or have alternative ideas/protocols in mind, please contact us and we will be happy to help where we can.
Sequencing on Illumina MiSeq
The MiSeq platform provides a flexible approach to next generation sequencing. The MiSeq provides;
- longer read lengths (up to 300bp, paired-end),
- reduced running times,
- dual-indexing supported for greater depth multiplexing of samples.
Below are the specifications for the MiSeq. These have been produced with the v3 Illumina consumables which are used routinely:
Read Length | Total Running Time | Approx. Output |
2 x 75bp | ~24hrs | 3Gb |
2 x 300bp | ~65hrs | 12Gb |
Number of Reads | |
Single Reads | 20M |
Paired End Reads | 40M (20M in each direction) |
If the run lengths above do not suit your needs or you would like any further information or help with your NGS experiment then please contact us. There are other read lengths available.
Please see the Genomics Pricing page for our costs.
Sequencing on Illumina NextSeq
The NextSeq platform offers a higher yield. We can offer this as a limited service in-house or outsource if appropriate. The NextSeq has two types of flow cell with run lengths determined by the user (up to 150bp paired-end);
- High-output - approx. 400 million reads/read-pairs per run.
- Mid-output - approx. 130 million reads/read-pairs per run.
If you require any further information (including costs) then please contact us.
Oxford Nanopore Technologies - MinION
We can assist you with you library preparation and provide sequencing using the ONT MinION platform. This is a new service and we are happy to hear more about researchers' experimental needs.
Sample Storage
We will endeavour to store remaining sample material or completed libraries for a period of 12 months before disposal.
Data Storage
We will endeavour to store data for a period of 12 months before deletion.
